I understand you can't really use a real fraternity's letters in a movie without their permission, but what about using an already created fake fraternity? Like Delta Tau Chi from Animal House? Would the screenwriters or production people have any kind of claim over a fake letter combination if the original movie wasn't mentioned?

I've noticed nobody reuses the fake fraternity letters but I'm not sure if there is a legal reason or artistic reason.

Unless the faked fraternity name is the subject of a trademark, there shouldn't be any problem using it, although you should probably search thoroughly to ensure that there isn't actually a fraternity with that name.
